The lake is of glacial origin and is located in the region of Lombardy in Italy, 45 kilometres north of Milan. Spread over an area of 146 kilometres, Lake Como is the third largest lake in Italy. The region offers an unparalleled opportunity for a lakeside retreat rich in historical, artistic and cultural values. The villas and the antique Romanesque churches transport you to a different world altogether. These villas are mostly encircled by admirable gardens full of lush sub-tropical plants. Source text - English A search of the available published and unpublished resources, including from the Abilify Maintena® Summary of Product Characteristics (SmPC), revealed the following information relevant to this request.
Generally, steady state is assumed to be reached in 4 to 5 times a drug’s half- life, following regular dosing (i.e. where the overall intake of a drug is in reasonably dynamic equilibrium with its elimination). In this case, for Abilify Maintena, steady state concentration is attained by the fourth dose, taking into account as stated in the Summary of Product Characteristics (SmPC) that:-
• aripiprazole’s absorption into the systemic circulation is slow and prolonged following Abilify Maintena administration due to low solubility of aripiprazole particles (average absorption half-life is 28 days);
• after every monthly Abilify Maintena injection of 400 mg, the mean aripiprazole terminal elimination half-life is 46.5 days, presumably due to absorption rate-limited kinetics;
• less than dose-proportional increases in aripiprazole and dehydro-aripiprazole (the active metabolite) concentrations and AUC parameters were observed after monthly Abilify Maintena injections of 400 mg. 1
